Pamela Isley was a botanist who was arrested for two accounts of murder, around the same time Joker was finding his footing as Gotham's public enemy nº1.

Isley killed her first victim through oleander poisoning, presumably mixed in with perfume. Her death was regarded by the G.C.P.D. as a mere allergic reaction, but Harleen Quinzel recognized the symptoms and tipped off the police signs of foul play. They later confirmed that it was indeed a premeditated murder.

The second victim was discovered alongside a string of Joker's spree killings. She had a naturally permanent smile on her face, different from other victims, who had their mouths tampered with coat hangers to achieve the same result. Lt. James Gordon suggested she was another Joker victim, but there were other inconsistencies that prevented the claim to become official, specially as Gordon's new appointed consultant Harleen Quinzel ran a toxicology test and found traces of strychnine in the victim's body, explaning how the victim's mouth contorted into a smile post-mortem. This pattern in poisoning confirmed the two victims were related. After Quinzel profiled the killer, the police eventually tracked down Pamela Isley's lab, which was full of raw material for the toxins they were looking for, leading to her arrest.
